Geboren als Sean Patrick Duke op 25 februari 1971 in Santa Monica. Zoon van Patty Duke. Toen zijn moeder in 1972 trouwde met John Astin adopteerde deze haar zoon. Patty Duke en John Astin kregen samen in 1973 Mackenzie Astin, Seans halfbroer. Sean Astin is bekend geworden door de Goonies, maar de meeste mensen zullen hem herkennen als de Hobbit Sam uit Lord of the Rings-trilogie.
